BizTalk Server 2016 supports Windows Server 2016, so in terms of BizTalk, it is possible.
In terms of SO, direct migration from Windows Server 2012 R2 to Windows Server 2016 Datacenter is supported.
So, I don’t find any reason not to be possible.
Thanks Sandro, i did it on a dev machine, post upgrade the BizTalk Admin MMC was broken, but after i did a BizTalk ‘Repair’ things seemed to be working as expected.
